How can I set up the shipping cost based on Fedex shipment rate?

I would like to create a shopping cart and I need to ship overnight and based
on rate. How can I create the shopping cart to charge the customers based on
how much they have purchased? I can get the Fedex calculator to let customer
to estimate the cost but how can I integrate to the payment shopping cart?
Please advise.
