Having problem changing the startup file

Version: 2004 Operating System: Mac OS X 10.6 (Snow Leopard) Processor: Intel Can someone give me a refresher on start up and commmand new workbook template thing? I want to change them both to 125% with large screen I have, but can't seem to do it. thanks!
