Out of Office Assistant Autoreply

Can I set Out of Office Assistant to reply every time an email received even
if it has responded to an email from that person already? We have issues
with people who are out for extended periods of time, e.g. maternity leave.

No (I'm presuming you use Exchange - always mention full details in here,
incl. your version of Outlook). OOF replies only once per sender to prevent
mail loops. It's really up to the sender to remember what they were told in
the OOF reply. Maybe you should grant someone else rights to the mailbox to
see if any new urgent items have come in.
