Can I create custom layouts in Publisher?

Of course you can! You can do just about anything you can imagine. I
guess we don't say that often enough around here. You DON'T NEED A
TEMPLATE for most things. Sketch your design and build it. It's very
easy! Write back if you have specific questions.

I have no problems personally doing layouts and making this nice robust
program do pretty much what I want. But I have other users in my workgroup
who do not have the inclination to learn everything that they should so that
they can completely utilize the program. They want to be able to click on a
layout icon and have it applied to the page they are working on. I've tried
it with Master Pages and it doesn't work.

That's not what you asked. You asked if you can create custom layouts. Yes,
you can. Since your people don't want to start from an existing template and
don't want to learn how to create their own templates, then it looks like
you'll have to create some sort of a template for them and teach them how to
use it.
