After updating Office 2000 with Office 2003, is it safe to remove the old
version from my hard drive to free up space?

If it was an Update, it should have removed the original as part of the
process.

It depends on the version of Office 2000 - some came with FrontPage and/or
Publisher which is not included on any Office 2003 CD and must be purchased
separately.

Publisher is certainly in Professional 2003 - as per MS's website!
