I mistakenly said no when I should have said yes to "do you want to save
changes". I worked all night on a newsletter, is there anyway to retrive my
unsaved changes. Thanks, I sure hope I can.

I doubt it... You might lucky and Publisher created a temp file. Type %temp% in
the address bar of Windows Explorer. Find any temporary file with the prefix
pub. Change the .tmp to .pub and try opening the file.

I've done that too (with various programs)
I think it's a "live and learn" thing.
