Lookup newbie question

Hi all, I'm trying to create an inventory program that will accept
information from an input screen and afterwards be able to upload the
information back onto my 400 box. Let me first say that I'm not a total
novice to Access but after this episode I'm starting to rethink it. So
here's what I'm trying to do:

I'm downloading all the items that I want to count but I need a field where
I can scan or type in the item number and type in the amount that was
counted. The problem I'm having is that for what I want to do it appears as
though I need to do a lookup but I'm totally unfamiliar with the process and
even more less familiar with forms.

I've downloaded the inventory template from MS site and I would like to
modify the template in order to get it to do what I want but it's not going
well and i'm seeking help.

"400 box"? IBM AS 400, to a table in a database on which you are linked?
Gee, we would probably need to have some details, including an idea of how
you have the data stored (table, table layout, data type) to give any
specific help.
